Computer System Validation and Electronic Records

Computer system validation establishes documented evidence that a computerised system used to generate, process, or store regulated data functions consistently and reliably in accordance with its intended use, encompassing verification of user requirements, functional specifications, installation qualification, operational qualification, and performance qualification. Validation extends beyond the software itself to encompass the surrounding process, including data backup and restoration procedures, user access control, and periodic review to confirm continued fitness for purpose throughout the operational life of the system.

Title 21 Code of Federal Regulations Part 11, issued by the United States Food and Drug Administration, establishes the criteria under which electronic records and electronic signatures are considered equivalent to paper records and handwritten signatures for regulatory purposes. Central to this regulation is the requirement for secure, computer-generated, time-stamped audit trails that independently record the date, time, and identity of the individual responsible for creation, modification, or deletion of electronic records, and that preserve the original data alongside any subsequent change. Effective audit trail review, conducted at a frequency commensurate with the criticality of the data and integrated into routine batch record review and periodic self-inspection, represents one of the most important practical controls for detecting and deterring data integrity breaches.
