Marker Compound Isolation

A marker compound is a chemically defined constituent, selected either for its known pharmacological activity or simply for its reliable, quantifiable presence, that serves as the analytical reference point for standardising an entire herbal extract. Isolation of a candidate marker compound follows the same column chromatography and preparative HPLC workflow, but with the specific objective of obtaining sufficient pure material — typically tens to hundreds of milligrams at greater than 95% purity, confirmed by HPLC and nuclear magnetic resonance spectroscopy — to serve as an in-house or certified reference standard against which every future production batch can be quantitatively compared. Well-established examples include curcumin from Curcuma longa, withanolide A from Withania somnifera, and andrographolide from Andrographis paniculata, each of which now functions as the primary quality-control marker for its respective botanical drug.
